Capsule (NRN: 04-5325): Omeprazole BP 20 mg (as enteric coated granules).
Pack size: Blister pack of 20 capsules.
It is very potent inhibitor of gastric acid output. Exerts its action by specific inhibition of H+/K+ ATPase enzyme system at the secretory surface of gastric parietal cell. It blocks the final step of acid production and is dose related and inhibits both basal and stimulated acid secretion.
Pharmacokinetics: It is a prodrug and is metabolised to active metabolite.
Onset of action: 2-5 hours.
Duration of action: 24 hours.
Duodenal and gastric ulcer, reflux or ulcerative oesophagitis, Zollinger-Ellison syndrome, NSAID induced ulcers.
Exclude gastric malignancy, renal and hepatic dysfunction.
Diazepam: Half life is increased.
Phenytoin: Increased plasma concentration.
Oral anticoagulants: May prolong the elimination of warfarin.
Nausea, constipation, diarrhoea, skin rashes, headache.
Dosage & Administration
Adults: Gastric ulcer: 20 mg once daily for 8 weeks; in severe cases increase to 40 mg once daily.
Duodenal ulcer: 20 mg once daily for 4 weeks
Zollinger Ellison Syndrome: 60 mg once daily.
Maintenance therapy: 20-120 mg adjusted to response; doses over 80 mg daily should be divided in 2 doses.
