Dosage Form, Composition & NAFDAC Registration Number (NRN)

Tablet (NRN:   ): Each uncoated tablet contains Metronidazole B.P. 200 mg.

Pack Size: 1000 tablets per jar. 100 tablets per jars. 10 blister of 10 tablets per box.

Syrup (NRN:   ): Each 5 mL (1 teaspoonful) contains Metronidazole Benzoate R P. eq. to Metronidazole RP 200 mg.

Pack Size: Bottle of 60 mL

Pharmacology

Action: Metronidazole is a broad spectrum antimicrobial and antiprotozoal drug, (being directly trichomonocidal). Selectively toxic to anerobic or microaerophilic microorganism and for anoxic and hypoxic cells.

Indications

A. Treatment and prophylaxis of anaerobic bacterial infections: 
* Bacterial vaginitis (nonspecific vaginites) * Ulcerative gingivitis * Pseudomembranous colitis. *Eradication of H.pylori in peptic ulcers 

B. Management of malodorous tumors and ulcers where there is anaerobics infections used as an adjunctant to radiotherapy of malignant neoplasms.

C. Treatment of Rosacea and of dracontiasis.

D. Treatment of infection due to R fragilis.

E. Prevention of post-operative anaerobic bacterial infection especially in patients undergoing abdominal and gynaecological surgery.

F. Treatment of parasitic protozoal infections: Amoebiasis, Giardiasis, Leishmaniasis, Trichomoniasis,Trypanosomiasis.

G. Drug of choice in mixed amoebicides.

Contra-indications

Precautions/Warnings

* Care should be exercised in treatment of patients with blood dyscrasias or with active disease of CNS

• Patients being treated for more than 10 days, should be monitored for any sign of peripheral neuropathy or CNS toxicity.

• Reduced doses should be given to patients with severe obstructive hepatic disease, alcoholic Cirrhosis or severe renal dysfunction.

• Use of Metronidazole should be avoided during pregnancy. Women taking Metronidazole should not breastfeed thin babies.

Interactions

Metronidazole has disulflrarn like effect, such that patients experience abdominal distress, vomiting, flushing or headaches on consumption of alcohol beverages during course of treatment. Psycoticslates may also occur during concurrent administration of Metronidazole and disulfiram.

Adverse Effects

Adverse effect are mainly dose related: Nausea, Unpleasant metallic taste, Headache, Anaeroxia, Vomitting, Diarrhoea, Dry mouth, Furred tongue, Glossitis, Stomatitis, Pseudomembranous colitis.

CNS side effects: Peripheral neuropathy, Weakness, Dizziness, Ataxia, Drowsiness, Changes in mood/mental state, insomnia, epileptic form seizures.

• Temporary Leucopenia.

• Skin rashes, pruritis

• Urethral discomfort and darkening of urine.

Dosage & Administration

TREATMENT ADULTS CHILDREN
AMOEBIASIS

Oral dose of 400 - 800 mg
3 times daily

for 5 - 10 days.

1 - 3 years 100 - 200 mg
(2.5 mL or teaspoon) 3 times daily
3 - 7 years 125 - 300 mg
(5 mL or 1 teaspoonful) 3 limes daily
7 - 10 years 200 - 400 mg
(5 - 10 mL or 1 - 2 teaspoonful) 3 times daily
GIARDIASIS

Oral dose of 2g once daily
in divided

(12.5 mL per day) in 3 divided doses for 3 days
